Soil nitrogen content, structure, and nitrogen cycling play a crucial role in tobacco growth quality, with different preceding crops having varying impacts on tobacco cultivation soil. This study conducted using field experiments, employed three treatments with different preceding crops, namely tobacco, barley, and rapeseed, to investigate the effects of different preceding crops on soil nitrogen structure and the expression levels of soil nitrogen cycling-related functional genes in tobacco cultivation soil. The results indicated that different preceding crops had varying effects on the content of different nitrogen forms in tobacco cultivation soil. Ammonium nitrogen and nitrate nitrogen were the two nitrogen forms which were most influenced by preceding crops, with the ammonium nitrogen content in soils following barley and rapeseed preceding crops increasing by 82.88% and 63.56%, respectively, compared to sole tobacco cultivation. The nitrate nitrogen content in tobacco cultivation soil was 26.97% higher following barley preceding crops and 24.39% higher following rapeseed preceding crops compared to sole tobacco cultivation. Simultaneously, different preceding crops also affected the expression levels of nitrogen cycling-related genes in tobacco cultivation soil. In the nitrification process, amoA was significantly impacted, with its expression reduced by 64.39% and 72.24% following barley and rapeseed preceding crops, respectively, compared to sole tobacco cultivation. In the denitrification process, except for the narG gene, all other genes were subjected to varying degrees of inhibition when preceded by barley and rapeseed crops. Correlation analysis between soil nitrogen structure and the expression levels of nitrogen cycling-related genes revealed that increased nitrogen levels suppressed the expression of Arch-amoA. Additionally, ammonium nitrogen strongly influenced the expression levels of most soil nitrogen cycling functional genes. In conclusion, preceding crops alter soil nitrogen structure, possibly due to changes in soil microorganisms, and different preceding crops modified the expression levels of nitrogen cycling-related genes in tobacco cultivation soil, consequently affecting the proportions of various nitrogen forms in the soil.

So far, coal, petroleum, and natural gas are still the most widely used fuels, and the emissions of SO2, NOX and particulate matter produced from their combustion have a serious influence on the air. Therefore, it is necessary to develop a clean fuel. In this study, the bulk curing barns were equipped with different fuel equipment, Barn A used traditional coal heating equipment; Barn B used biomass briquettes fuel (BBF) integrated heating equipment; Barn C equipped with alcohol-based fuel (ABF) heating equipment. The temperature of the outer surface of the heating equipment, the exhaust gas of the chimney, and the curing heat efficiency and energy consumption were analyzed. Compared with the barn BBF and barn coal, the barn ABF can meet the flue-cured tobacco curing highest temperature requirements of 68 °C, the accuracy of the target dry bulb temperature (DBT) curve during the curing of flue-cured tobacco was 93.4%. At the same time, during ABF combustion, the emissions of CO2 and CO were 40.82% and 0.19%, respectively. However, no emissions of NOX, SO2, and H2S were detected in the chimney exhaust. Compared with the barn BBF and barn coal, the thermal efficiency of barn ABF heating equipment in the barn was increased by 44.78% and 86.28%, respectively. Additionally, the coast per kilogram of dry tobacco was reduced by 19.44% and 45.28%, respectively. Therefore, compared to barn coal and barn BBF, the barn ABF can control temperature changes more accurately, and shows an obvious advantage in environmental protection and heat utilization efficiency.

Modern gas chromatography-mass spectrometry (GC-MS) is the workhorse for the high-throughput profiling of volatile compounds in complex samples. It can produce a considerable amount of two-dimensional data, and automatic methods are required to distill chemical information from raw GC-MS data efficiently. In this study, we proposed an Automatic Resolution method (AutoRes) based on pseudo-Siamese convolutional neural networks (pSCNN) to extract the meaningful features swamped by the noises, baseline drifts, retention time shifts, and overlapped peaks. Two pSCNN models were trained with 400,000 augmented spectral pairs, respectively. They can predict the selective region (pSCNN1) and elution region (pSCNN2) of compounds in an untargeted manner. The accuracies of the pSCNN1 model and the pSCNN2 model on their test sets are 99.9% and 92.6%, respectively. Then, the chromatographic profile of each component was automatically resolved by full rank resolution (FRR) based on the predicted regions by these models. The performance of AutoRes was evaluated on the simulated and plant essential oil datasets. Compared to AMDIS and MZmine, AutoRes resolves more reasonable mass spectra, chromatograms, and peak areas to identify and quantify compounds. The average match scores of AutoRes (925 and 936) outperformed AMDIS (909 and 925) and MZmine (888 and 916) when resolving mass spectra from overlapped peaks on the Set Ⅰ and Set Ⅱ of plant essential oil dataset and matching them against the NIST17 library. It extracted peak areas and mass spectra automatically from 10 GC-MS files of plant essential oils, and the entire process was completed in 8 min without any prior information or manual intervention. The first-cured tobacco contains macromolecular substances with negative impacts on tobacco products quality, and must be aged and fermented to mitigate their effects on the tobacco products quality. However, the natural fermentation takes a longer cycle with large coverage area and low economic efficiency. Microbial fermentation is a method to improve tobacco quality. The change of chemical composition of tobacco during the fermentation is often correlated with shapes of tobacco. This study aimed to investigate the effects of tobacco microorganisms on the quality of different shapes of tobacco. Specifically, Bacillus subtilis B1 and Cytobacillus oceanisediminis C4 with high protease, amylase, and cellulase were isolated from the first-cured tobacco, followed by using them for solid-state fermentation of tobacco powder (TP) and tobacco leaves (TL). Results showed that strains B1 and C4 could significantly improve the sensory quality of TP, enabling it to outperform TL in overall texture and skeleton of tobacco products during cigarette smoking. Compared with the control, microbial fermentation could increase reducing sugar; regulate protein, starch, and cellulose, reduce nicotine, improve total aroma substances, and enable the surface of fermented TP and TL to be more loose, wrinkled, and porous. Microbial community analysis indicated that strains B1 and C4 could change the native structure of microbial community in TP and TL. LEfSe analysis revealed that the potential key biomarkers in TP and TL were Bacilli, Pseudonocardia, Pantoea, and Jeotgalicoccus, which may have cooperative effects with other microbial taxa in improving tobacco quality. This study provides a theoretical basis for improving tobacco fermentation process for better cigarettes quality.

Organic fertilizer is effective in improving soil quality, and promoting crop growth. Combined organic and inorganic fertilization has been proved as a more favorable way to tobacco yield and quality. However, the mechanisms underlying tobacco yield and quality under combinations of different organic and inorganic fertilizer remain unclear. We conducted a 12-year tobacco (Nicotiana tabacum L.)-maize crop rotation field experiment in Yanhe experimental station, China to examine the yields and qualities of tobacco, soil nutrients, and extracellular enzyme activities associated with carbon, nitrogen, and phosphorus cycles in response to different fertilization treatments. Five fertilization treatments (no fertilization; 75 kg N fertilizer ha-1; 450 kg oil cake ha-1 + 75 kg N fertilizer ha-1; 15,000 kg pig dung ha-1 + 60 kg N fertilizer ha-1; 3,000 kg straw ha-1 + 75 kg N fertilizer ha-1) were applied to tobacco while maize was fertilized with inorganic compound fertilizers. After 12 years of tobacco-maize rotation, the results showed that organic fertilizer additions elevate tobacco yield and quality, and the soil extracellular enzymes activities. Gram-negative bacteria, actinomycetes, and total soil microbial biomass were increased by organic fertilizer additions, both plant-based (oil cake and straw) and animal-based (pig dung) organics. The levels of soil organic matter, total organic carbon, total phosphorus and available phosphorus are higher in pig dung addition treatment than oil cake and straw additions. By variance analysis with respect to fertilization treatments, organic sources differentially affected the activities of diverse soil enzymes. The redundancy analysis gave that yield and quality of tobacco leaves (upper, middle, and lower leaves) positively related to soil extracellular enzyme activities. Based on analysis of yield and quality of tobacco leaves with extracellular enzyme activities and soil nutrients, it is suggested animal-based organic fertilizer, thus pig dung, should be used in combining with chemical fertilizers to improve the quality of tobacco and soil nutrients.

BACKGROUND: Weather change in high-altitude areas subjects mature tobacco (Nicotiana tabacum L.) to cold stress, which damages tobacco leaf yield and quality. A brupt diurnal temperature differences (the daily temperature dropping more than 20 °C) along with rainfall in tobacco-growing areas at an altitude above 2450 m, caused cold stress to field-grown tobacco. RESULTS: After the flue-cured tobacco suffered cold stress in the field, the surface color of tobacco leaves changed and obvious large browning areas were appeared, and the curing availability was extremely poor. Further research found the quality of fresh tobacco leaves, the content of key chemical components, and the production quality were greatly reduced by cold stress. We hypothesize that cold stress in high altitude environments destroyed the antioxidant enzyme system of mature flue-cured tobacco. Therefore, the quality of fresh tobacco leaves, the content of key chemical components, and the production quality were greatly reduced by cold stress. CONCLUSION: This study confirmed that cold stress in high-altitude tobacco areas was the main reason for the browning of tobacco leaves during the tobacco curing process. This adverse environment seriously damaged the quality of tobacco leaves, but can be mitigated by pay attention to the weather forecast and pick tobacco leaves in advance.

BACKGROUND: The leaf age for harvesting flue-cured tobacco leaves is closely related to the quality of tobacco leaves, so an appropriate leaf age for harvesting is important for improving yield and quality of flue-cured tobacco, however, at present, there are few studies on effects of leaf age on physiological and biochemical changes during flue-curing and there is no clear standard of proper leaf ages for harvesting in production. RESULTS: In the Yunnan tobacco-growing area, an experiment was carried from 2016 to 2017 and different leaf ages were set. The results demonstrate that leaf age has a significant on tissue cell gap, leaf age and flue-curing stages exert significant effects on upper epidermis, palisade and spongy tissue, and leaf thickness of tobacco leaves. The thicknesses of upper and lower epidermis as well as palisade and spongy tissues at different ages show an approximately W-shaped change trend during flue-curing. With the advance of flue-curing stages, contents of starch, chlorophyll, carotenoid, and water in tobacco leaves at different leaf ages decrease, while polyphenol and malondialdehyde (MDA) contents increase. The older the leaf, the faster the chlorophyll, carotenoid, and water contents reduce, while the faster the polyphenol and MDA content rise during flue-curing. The flue-cured tobacco leaves at 116 DAT (days after transplanting) show the highest contents of total nitrogen and nicotine, followed by 123 DAT and those at 130 DAT are the lowest; however, the contents of total sugar and reducing sugar demonstrate a contrary tendency, and the starch content at 116 DAT is much lower than those in the other two treatments. The proportion of superior tobacco, average price, yield, and output value of upper tobacco leaves at different leaf ages are the highest at 123 DAT. The highest sensory evaluation score is found at 123 DAT, while that at 130 DAT is significantly lower in comparison with the other two treatments. CONCLUSIONS: Tobacco leaves harvested at 123 DAT are mature and exhibit a low degree of membrane lipid peroxidation, moderate chemical compositions, and high economic value. 123 DAT improves availability of tobacco leaves.
